چکیده

The goal of the following research project was to perform a three-dimensional reconstruction of selected CDs and associated AVR as they ascend through the IM/OM border, and determine whether AVR in this region coalesce to form giant vascular bundles, or whether they do so in a more dispersed pattern, interacting with components of the interbundle regions. The previously accepted model of vascular architecture in this region suggests that as AVR ascend through the IM/OM border, they coalesce to form dense vascular bundles (Figure 5, Pallone). Previous research performed in this lab, along with current three-dimensional reconstructions suggest that AVR are not confined to dense vascular bundles as they enter the OM. Instead, we hypothesize that as intracluster AVR ascend through the region of interest, they do so in a more dispersed pattern throughout the inner stripe of the OM. Analysis of the following experiment utilized methods including immunohistochemical preparation of the tissue samples, epifluorescent microscopy, and three-dimensional reconstructions of AVR through the region of interest. The three-dimensional reconstruction of three CDs, and fiveassociated AVR resulted in two AVR joining the vascular bundle, while the other three were found to ascend through the interbundle regions of the OM.

Isolation and perfusion of rat inner medullary vasa recta.

Outer medullary isolated descending vasa recta have proven to be experimentally tractable, and consequently much has been learned about outer medullary vasa recta endothelial transport, pericyte contractile mechanisms, and tubulovascular interactions. In contrast, inner medullary vasa recta have never been isolated from any species, and therefore isolated vasa recta function has never been subj...

Architecture of vasa recta in the renal inner medulla of the desert rodent Dipodomys merriami: potential impact on the urine concentrating mechanism.

We hypothesize that the inner medulla of the kangaroo rat Dipodomys merriami, a desert rodent that concentrates its urine to over 6,000 mosmol/kg H(2)O, provides unique examples of architectural features necessary for production of highly concentrated urine. To investigate this architecture, inner medullary vascular segments in the outer inner medulla were assessed with immunofluorescence and d...

Architecture of interstitial nodal spaces in the rodent renal inner medulla.

Every collecting duct (CD) of the rat inner medulla is uniformly surrounded by about four abutting ascending vasa recta (AVR) running parallel to it. One or two ascending thin limbs (ATLs) lie between and parallel to each abutting AVR pair, opposite the CD. These structures form boundaries of axially running interstitial compartments. Viewed in transverse sections, these compartments appear as ...

Hydraulic and oncotic pressure measurements in inner medulla of mammalian kidney.

The vasa recta are thought to play an important role in the transfer of water andsolutes within the renal medulla. Hydraulic pressures were measured in vasa recta onthe surface of the exposed papilla in young Munich Wistar rats, and blood was collected from these microvessels for determination of total protein concentration and calculation of colloid oncotic pressure. In descending vasa recta...

Relation between vasa recta blood flow and renal interstitial hydrostatic pressure during pressure natriuresis.

Pressure natriuresis may be mediated through increases in inner medullary vasa recta blood flow (QVR). By means of acute renal decapsulation to prevent increases in renal interstitial hydrostatic pressure (RIHP), the effect of increases in QVR in the presence and absence of changes in RIHP in the natriuretic and diuretic responses to increases in renal perfusion pressure (RPP) was evaluated. Bl...
